Koena Mitra was born in Kolkata, West Bengal to middle class Bengali family on 7th January 1984. Her father Vishwanath Mitra is Charter Accountant and mother Krishna Mitra was a school teacher. She went to South Point High School, Kolkata and completed her graduation at Lady Brabourne College, Kolkata. She is a post-graduate in psychology. After deciding that she wanted to pursue acting and modeling as career she joined Lee Strasberg Theatre and Film Institute, Los Angeles.

Career
Her first screen appearance was in a item number Khullam Khulla of Road (2002) followed by another appearance in item number Koduva Meesai of a Tamil film Dhool in 2003. But her fortune flipped the coin after her another item song Saki Saki of Musafir in 2004, this song made her overnight star and she get a nickname The Saki Girl. Later, Koena did many Bollywood films like Ek Khiladi Ek Haseena, Insan, Apna Sapna Money Money, Hey Babyy.
Koena made her first appearance in television as a guest in dance reality show Jhalak Dikhla Jaa season 3 aired on Sony TV. She participated in Bigg Boss season 13 as contestant afterwards but she can not hold the show for a long go and eliminated after just 14 days.
